ABSTRACT

Objective:To analyze the influencing factors of delayed nausea and vomiting in patients with primary liver cancer after transarterial chemoembolization based on Logistic regression model and decision tree model.Methods:This was a cross-sectional study. A total of 236 patients with primary liver cancer after transarterial chemoembolization in The Second Affiliated Hospital of Air Force Military Medical University from March 2021 to June 2022 were conveniently selected as the research subjects. The factors related to delayed nausea and vomiting were collected, and Logistic regression and decision tree models were established, respectively, and the differences between the two models were compared.Results:The incidence of delayed nausea and vomiting of patients with primary liver cancer after transarterial chemoembolization was 45.34% (107/236). Logistic regression model showed that age, anxiety, sleep disorder, emetic risk level of chemotherapeutic drugs, embolic agent type, and pain 24 hours after surgery were the influencing factors of delayed nausea and vomiting in patients with primary liver cancer after transarterial chemoembolization(all P<0.05). Decision tree model showed that age, sleep disorder, emetic risk level of chemotherapeutic drugs, embolic agent type, and pain 24 hours after surgery were the influencing factors of delayed nausea and vomiting in patients with primary liver cancer after transarterial chemoembolization (all P<0.05). The classification accuracy rates of Logistic regression, decision tree model and combined diagnosis of two models were 72.9%, 71.2% and 72.0% respectively; the areas under the ROC curve were 0.778, 0.781 and 0.806 respectively, with no significant difference (all P>0.05). Conclusions:The analysis results of Logistic regression and decision tree model on the influencing factors of delayed nausea and vomiting in patients with primary liver cancer after transarterial chemoembolization are highly consistent, which can be combined to provide a more comprehensive reference for the evaluation and intervention of medical staff.

@#Abstract:Objective To carry out serological analysis of varicella⁃zoster virus(VZV)IgG antibody level in healthy people aged 1 ~ 30 years in Liaoning Province. Methods In October 2020,3~5 mL venous blood samples were collected from 617 healthy people aged 1~30 years selected from six counties and districts in Shenyang,Fuxin and Dandong of Liaoning Province by stratified random sampling method,of which serum samples were collected and determined for VZV IgG antibody level by ELISA. The positive rate of serum antibody and geometric mean concentration(GMC)of antibody were calculated and compared. Results Among 617 serum samples,302 samples were positive for VZV IgG antibody,the positive rate was 48. 947%,and the GMC was 112. 772 mIU/mL. The positive rate of VZV IgG antibody was 29. 670%~75. 789% and the GMC was 45. 508~366. 559 mIU/mL in healthy people of various ages. Both of the antibody positive rate(χ2 = 67. 104, P < 0. 001)and GMC(F = 20. 685,P < 0. 001)showed significant differences. The positive rates of VZV IgG antibody in male and female were 44. 817% and 53. 633% respectively,which showed significant difference(χ2 = 4. 779,P = 0. 029), while the GMCs were 96. 983 and 133. 829 mIU/mL respectively(t = -1. 958,P = 0. 051)with no significant difference. The positive rates of VZV IgG antibody of healthy people in Shenyang,Fuxin and Dandong of Liaoning Province were 55. 224%,40. 201% and 51. 152% respectively with significant differences(χ2 = 9. 683,P = 0. 008),of which the positive rate of FuxinwassignificantlylowerthanthoseofShenyangandDandong(χ2 =9. 046and5. 013,P =0. 003and0. 025,respectively); While the GMCs were 133. 523,85. 953 and 123. 713 mIU/mL respectively with no significant difference(F = 0. 514, P = 0. 598). Among 617 serum samples,54 sampleswere suspicious,which remained within the criticalrange afterre⁃examina⁃ tion,while the gap between positive rate and the total percentage of positive and suspicious results gradually decreased with the increase of age,indicating that the immunity to varicella gradually increased with the increase of age. Conclusion The VZV⁃IgG antibody level of healthy people aged 1~30 years in Liaoning Province increased gradually with age,while the overall level was low. To control the spread of varicella virus,it is recommended to increase varicella vaccine coverage in vulnerable areas and susceptible population to build VZV immune barrier.

@#Abstract: Objective By collecting and sorting the information of varicella cases reported in Liaoning Province from 2006 to 2021, the epidemiological characteristics were analyzed, and the monthly incidence data were predicted, so as to explore the prevention and control strategy of varicella disease in Liaoning Province. Methods By collecting the characteristic information of varicella cases in Liaoning Province, epidemiological analysis was carried out on the regional, population, and temporal characteristics of varicella incidence. The monthly incidence data of varicella were fitted with Eviews software, seasonal ARIMA model was used for modeling, and models were selected according to SC and AIC. After modeling, the model was used to predict the incidence data in 2022. Results The incidence rate of varicella in Liaoning Province has increased in recent years. The onset time was "bimodal distribution", with the main peak occurring from November to January of the next year and the secondary peak occurring from May to June. Since 2019, the onset age has shifted backward. From the original 0-<10 age group with the highest incidence rate, it shifted to the 10-<20 age group with the highest incidence rate. From 2006 to 2021, the incidence of varicella mainly concentrated in people aged 0 to <40 years old, and the incidence rate of the population over 40 years old showed a cliff-like decline. The incidence of chickenpox was higher in the central region of Liaoning Province, such as Shenyang, Dalian, Anshan and Panjin, and relatively low in Huludao, Jinzhou, Fuxin and Liaoyang. The distribution of the population was mainly students, followed by kindergartens and scattered children. ARIMA model of monthly incidence data was established by software as ARIMA (1, 0, 1) (1, 1, 1)12. Conclusions The incidence rate of varicella in Liaoning Province has been rising in recent years. The incidence is obviously seasonal, and the age group of the affected population has moved backward. It is predicted that the incidence will continue to increase in 2022. The prevention and control of varicella should still be the current key work. In order to reduce the population incidence rate, two-dose vaccination strategies should be vigorously promoted the implementation of the, and the inclusion of varicella vaccine in the immunization program should be achieved as soon as possible.

@#Objective To analyze the research status and hotspot of occupational methanol poisoning at home and abroad. Methods , The China National Knowledge Resource Database Wanfang Data Knowledge Service Platform and Web of Science were used as the data sources. The relevant literatures on occupational methanol poisoning published in domestic and foreign , Results journals up to June 30 2021 were searched. The bibliometrics was used to analyzed the literatures. A total of 255 literatures were included in analysis. There were 187 Chinese articles and 68 English articles. Most of Chinese articles were , , published from 2001 to 2005 with an average of 26.7 literatures per five years until June 2021. Among them 72 literatures ( ), , were published in core journals 38.5% and 176 authors from 27 provinces autonomous regions and municipality directly , under the central government published relevant literatures. The research contents mainly focused on the classification , , poisoning mode clinical manifestations visual impairment and poisoning prevention and treatment of occupational methanol - , poisoning. Most of the English literatures were published in 2016 2020 with an average of 4.9 articles per five years until June , ( ), 2021. Among them 36 were published in SCI journals 52.9% and 57 authors from 11 countries published relevant , , , literatures. The research contents mainly focused on the clinical diagnosis drug treatment intoxication mechanism visual Conclusion sequelae and brain injury of occupational methanol poisoning. The research on occupational methanol poisoning , , , mainly focuses on clinical diagnosis clinical manifestations treatment and prognosis and pathogenesis. The focus of relevant research at home and abroad is different.

ABSTRACT

Objective:To explore the influencing factors of disability acceptance in patients with spinal cord injury, and analyze the correlation between alexithymia and disability acceptance.Methods:The convenience sampling method was used to select 276 patients with spinal cord injury admitted to Affiliated Hospital of Jining Medical College from October 2018 to June 2020 as the research objects. The general information questionnaire, the Toronto Alexitymia Scale (TAS-20), and the Acceptance of Disability Scale (ADS) was used to conduct the survey, Pearson correlation was used to analyze the correlation between alexitymia and disability acceptance in patients with spinal cord injury, hierarchical regression analysis was used to analyze the factors that affect the disability acceptance of patients with spinal cord injury.Results:The total score of ADS of 276 patients with spinal cord injury was (172.68 ± 37.72) points, and the total score of TAS-20 was (55.60 ± 9.49) points. The results of Pearson correlation analysis showed that all dimensions and total score of TAS-20 were negatively correlated with all dimensions and total score of ADS ( r values were -0.821--0.101, all P<0.05). The results of hierarchical regression analysis showed that gender, course of disease, post-injury working status were the risk factors of disability acceptance ( P<0.05), and alexithymia was the protective factor of disability acceptance( P<0.05), gender, course of disease, post-injury working status and alexithymia level could jointly explain the 22.1% variation in disability acceptance in patients with spinal cord injury. Conclusions:The disability acceptance of patients with spinal cord injury is at a moderate level, and alexitymia is negatively correlated with disability acceptance. alexitymia, gender, course of disease, and post-injury work status are the influencing factors of disability acceptance of patients with spinal cord injury.

ABSTRACT

Objective:To explore the correlation between lower gastrointestinal tract symptoms and diabetic retinopathy and lower extremity artery disease in patients with type 2 diabetic mellitus.Methods:The clinical data of 128 patients with type 2 diabetic mellitus from August 2016 to March 2018 in Central Hospital of Shengli OilField were retrospectively analyzed. The general clinical data and laboratory indexes were recorded. The score was evaluated by lower gastrointestinal symptoms rating scale (LGSRS). Patients with the LGSRS score ≥6 scores were as positive group, and those with LGSRS score<6 scores were as negative group. Fundus photography and ultrasonic scanning of lower extremity artery disease were performed for each patient. The correlation between LGSRS score and diabetic retinopathy, lower extremity artery disease was analyzed by Logistic regression.Results:Positive group had 66 cases, and negative group had 62 cases. The age, duration of diabetic, LGSRS score and incidence of diabetic retinopathy in positive group were significantly higher than those in negative group: (64.24 ± 10.43) years vs. (59.57 ± 12.50) years, (14.67 ± 1.28) years vs. (10.98 ± 1.25) years, (8.65 ± 2.38) scores vs. (2.77 ± 1.49) scores and 98.48% (65/66) vs. 69.35% (43/62), and there were statistical differences ( P<0.05 or<0.01). There were no statistical difference in gender, body mass index, glycated hemoglobin, fasting blood glucose, 2 h postprandial blood glucose, triglyceride, total cholesterol and incidence of diabetic retinopathy between 2 groups ( P>0.05). After adjusting for age, duration of diabetic, glycated hemoglobin, fasting blood glucose, triglyceride and total cholesterol, the LGSRS score showed positive correlation with diabetic lower extremity artery disease ( OR = 1.174, 95% CI 1.065 to 1.470, P = 0.001), but there was no correlation between LGSRS score and diabetic retinopathy ( P = 0.313). Conclusions:In patients with type 2 diabetes mellitus, lower gastrointestinal tract symptoms has positive correlation with diabetic lower extremity artery disease, and no correlation with diabetic retinopathy. It suggests that there may be a common pathological mechanism between lower gastrointestinal tract symptoms and diabetic lower extremity artery disease.

ABSTRACT

Objective:To investigate the expressions of miR-20a-5p and lysine (K) demethylase 6B (KDM6B) in osteosarcoma tissues and the effects of miR-20a-5p targeting KDM6B on the proliferation, migration and invasion of osteosarcoma cells and tumor growth.Methods:The clinicopathological and paracancerous tissues of 20 patients with osteosarcoma admitted to the First Affiliated Hospital of Chinese Medical University from January 2017 to March 2019 were collected. Quantitative real-time PCR (qRT-PCR) was used to detect the expression levels of miR-20a-5p and KDM6B mRNA in tissues. The osteosarcoma MG63 cells were divided into control group, mimic NC group, miR-20a-5p mimic group, and NC+ empty vector group, miR-20a-5p+ empty vector group, miR-20a-5p+ KDM6B group. The expression levels of miR-20a-5p and KDM6B mRNA of all groups were detected by qRT-PCR. Western blotting was used to detect the expression level of KDM6B. CCK-8 assay, cell scratch test and Transwell test were used to detect cell proliferation, migration and invasion ability. According to the random number table method, nude mice were divided into NC+ empty vector group, miR-20a-5p+ empty vector group and miR-20a-5p+ KDM6B group, with 5 mice in each group. Tumor growth ability was detected by tumor xenograft nude mouse models.Results:The relative expression level of miR-20a-5p mRNA in osteosarcoma tissues was 0.55±0.27, and that in paracancerous tissues was 1.22±0.28, with a statistically significant difference ( t=7.701, P<0.001). The relative expression level of KDM6B mRNA in osteosarcoma tissues was 1.66±0.19, and that in paracancerous tissues was 1.00±0.15, with a statistically significant difference ( t=12.219, P<0.001). After transfection of miR-20a-5p, KDM6B mRNA and protein expression levels decreased with the increase of miR-20a-5p expression level. After miR-20a-5p transfection for 48 h, the cell proliferation abilities of the blank control group, mimic NC group and miR-20a-5p mimic group were 0.83±0.04, 0.81±0.03 and 0.52±0.01 ( F=89.655, P<0.001), compared with the blank control group and mimic NC group, the cell proliferation ability was significantly inhibited in the miR-20a-5p mimic group (both P<0.001). The cell proliferation abilities of NC+ empty vector group, miR-20a-5p+ empty vector group and miR-20a-5p+ KDM6B group were 0.83±0.05, 0.52±0.01 and 0.67±0.05 ( F=43.919, P<0.001), compared with the NC+ empty vector group, the cell proliferation ability was significantly inhibited in the miR-20a-5p+ empty vector group ( P<0.001); compared with the miR-20a-5p+ empty vector group, the cell proliferation ability of miR-20a-5p+ KDM6B group increased significantly ( P<0.001). The scratch healing rates of the blank control group, mimic NC group and miR-20a-5p mimic group were (32.51±2.73)%, (30.26±3.22)% and (13.52±1.77)% ( F=46.314, P<0.001), compared with the control group and the mimic NC group, the scratch healing rate of the miR-20a-5p mimic group was significantly decreased (both P<0.001). The scratch healing rates of NC+ empty vector group, miR-20a-5p+ empty vector group and miR-20a-5p+ KDM6B group were (31.34±3.11)%, (12.15±1.64)% and (28.93±2.89)% ( F=47.511, P<0.001), compared with the NC+ empty vector group, the scratch healing rate of the miR-20a-5p+ empty vector group was significantly decreased ( P<0.001); compared with the miR-20a-5p+ empty vector group, the scratch healing rate of miR-20a-5p+ KDM6B group was significantly increased ( P=0.001). The numbers of transmembrane cells in the blank control group, mimic NC group and miR-20a-5p mimic group were 114±16, 108±11 and 42±6 ( F=36.282, P<0.001), compared with the control group and mimic NC group, the number of transmembrane cells of the miR-20a-5p mimic group was significantly decreased (both P<0.001). The numbers of transmembrane cells in the NC+ empty vector group, miR-20a-5p+ empty vector group and miR-20a-5p+ KDM6B group was 143±11, 39±4 and 139±12 ( F=112.120, P<0.001), compared with the NC+ empty vector group, the number of transmembrane cells of the miR-20a-5p+ empty vector group was significantly decreased ( P<0.001); compared with the miR-20a-5p+ empty vector group, the number of transmembrane cells of the miR-20a-5p+ KDM6B group was increased significantly ( P<0.001). The tumor volumes of mice for 21 d in the NC+ empty vector group, miR-20a-5p+ empty vector group and miR-20a-5p+ KDM6B group were (1 667.50±250.40) mm 3, (129.20±21.00) mm 3 and (775.41±77.51) mm 3 respectively, with a statistically significant difference ( F=77.651, P<0.001). The tumor weights of the 3 groups were (1.35±0.18) g, (0.12±0.01) g and (0.61±0.03) g respectively, with a statistically significant difference ( F=104.191, P<0.001). Conclusion:The expression of miR-20a-5p is significantly decreased in osteosarcoma tissues, and the expression of KDM6B is significantly increased in osteosarcoma tissues. Overexpression of miR-20a-5p may inhibit the proliferation, migration and invasion of osteosarcoma cells and tumor growth by targeting to reduce the expression of KDM6B.

ABSTRACT

OBJECTIVE: To study the association between superior mesenteric artery hemodynamic indexes and scores of lower gastrointestinal symptoms rating scales(LGSRS) in patients with type 2 diabetic mellitus. METHODS: Totally 142 inpatients with type 2 diabetes with average age of 58.76±12.32 yrs were enrolled, who were treated from August 2016 to March 2018. The history, gender, age,course and BMI were recorded, and fasting blood glucose(FBG), glycosylated hemoglobin(HbA1c), 2-hour postprandial blood glucose(PBG), total cholesterol(TC), triglyceride(TG), urine ACR and LGSRS were determined. Ultrasonic scanning of mesenteric artery was performed for hemodynamic indexes, including artery inner diameter(ID), peak systolic velocity(PSV), end-diastolic velocity(EDV), and resistance index(RI)at starting part,first level branch, and second level branch from root of the superior mesenteric artery(SMA).Patients were divided into 2 groups according to their LGSRS, 74 patients with LGSRS≥6 were in positive group, and 68 patients with LGSRS0.05), but the age and DD were significantly higher in positive group than in control group(P0.05). 3. There were no significant difference between positive group and control group in ID at starting part and first level branch of SMA, while ID at second level branch was significantly increased in positive group compared with control group [(3.83±0.85)mm vs.(3.53±0.90)mm, P<0.05)].4. RI at first(0.816±0.059 vs 0.842±0.063,P<0.05) and second level branch(0.813±0.076 vs 0.845±0.073, P<0.05) and PSV at first level branch[(110.89±46.89)cm/s vs(95.72±36.59)cm/s,P<0.05] were significantly high in positive group; there were no difference in other hemodynamic indexes between the groups. 5.Adjusted by age,DD,glycemic and lipidemic profile,Logistic regression showed that ID at first(RR=2.092,95%CI 1.080-4.050,P=0.029) and second level branch(RR=0.491,95%CI 0.252-0.955,P=0.36) and EDV at second level branch(RR=0.897,95%CI 0.824-0.976,P=0.012) were independent factors influencing LGSRS(P<0.05). CONCLUSION: Ultrosonic hemodynamic abnormalities in the superior mesenteric artery might be important factor in development of lower gastrointestinal tract symptoms in patients with type 2 diabetes.

ABSTRACT

Abstract Streptomycetes remain as one of the important sources for bioactive products. Isolated from the mangrove forest, Streptomyces gilvigriseus MUSC 26T was previously characterised as a novel streptomycete. The high quality draft genome of MUSC 26T contained 5,213,277 bp with G + C content of 73.0%. Through genome mining, several gene clusters associated with secondary metabolites production were revealed in the genome of MUSC 26T. These findings call for further investigations into the potential exploitation of the strain for production of pharmaceutically important compounds.

ABSTRACT

ABSTRACT As the largest genus in Actinobacteria family, Streptomyces species have the ability to synthesize numerous compounds of diverse structures with bioactivities. Streptomyces mangrovisoli MUSC 149T was previously isolated as a novel streptomycete from mangrove forest in east coast of Peninsular Malaysia. The high quality draft genome of MUSC 149T comprises 9,165,825 bp with G + C content of 72.5%. Through bioinformatics analysis, 21 gene clusters identified in the genome were associated with the production of bioactive secondary metabolites. The presence of these biosynthetic gene clusters in MUSC 149T suggests the potential exploitation of the strain for production of medically important compounds.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of early rehabilitation intervention on the incidences of extrauterine growth retardation (EUGR) and early diseases in preterm infants.</p><p><b>METHODS</b>The appropriate-for-gestational-age preterm infants with a gestational age of <34 weeks and a birth weight of 1 000 to <2 000 g who were admitted to the neonatal intensive care unit (NICU) within 24 hours after birth were enrolled in a prospective randomized controlled trial. These infants were randomly divided into rehabilitation intervention group and control group. The infants in the rehabilitation intervention group were given early rehabilitation after their vital signs became stable, including oral sensory and muscle strength training and pressure touching of the head, chest, abdomen, extremities, hands, and feet. The primary outcome measures were the time to independent oral feeding, length of hospital stay, and incidence rate of EUGR. The secondary outcome measures were the incidence rates of related diseases in preterm infants, such as apnea, feeding intolerance, and sepsis.</p><p><b>RESULTS</b>A total of 97 preterm infants who met the inclusion criteria and had complete data were enrolled, with 48 in the control group and 49 in the rehabilitation intervention group. The rehabilitation intervention group had a shorter time to independent oral feeding than the control group (P<0.05). Compared with the control group, the rehabilitation intervention group had a shorter length of hospital stay and a lower corrected gestational age at discharge (P<0.05), as well as a lower incidence rate of EUGR (P<0.05). The rehabilitation intervention group ONCLUSIONS: Early rehabilitation intervention for preterm infants in the NICU may reduce the incidence rates of apnea, feeding intolerance, and EUGR and help them to achieve independent oral feeding early.</p>

Objective:To evaluate the bonding ability of one representative self-etch adhesive system by applying the micro-shear bond strength test method with different concentrations of carbodiimide [1-ethyl3-(3-dimethylaminopropyl) carbodiimide,EDC] ethanol solution pretreatment.Methods:Extracted sound human third molars were collected from patients ranging 18 to 40 years.In the study,80 molars were first sectioned to prepare a flat middle coronal dentin surface and then were randomly divided into 5 groups (n =16) according to without/with different surface treatments [blank control;80% (volume fraction) ethanol control;EDC ethanol solution of three concentrations (0.01 mol/L,0.3 mol/L and 0.5 mol/L)].Each specimen underwent a micro-shear bond strength test and failure mode observation.The data collected were subjected to statistical analysis using one-way ANOVA and post hoc Tukey's test to analyze the difference of the micro-shear bond strength,and chi-square test/Fisher's exact test for the failure mode frequency of the micro-shear bond strength test specimens at a significance level of P =0.05.Results:The micro-shear bond strength of the three concentration EDC treatments were (35.29 ± 8.97) MPa (0.01 mol/L EDC treatment group),(40.24 ±9.68) MPa (0.3 mol/L EDC treatment group),(37.38 ±9.66) MPa (0.5 mol/L EDC treatment group) separately;and that of the 80% ethanol group was (37.49 ± 7.76) MPa.All micro-shear bond strength value of the above four groups was statistically higher than that of the blank control group [(33.81 ± 7.98) MPa].The immediate microshear bond strength and failure mode was of no statistically significant difference among all the groups.It was noticed that the immediate micro-shear bond strength of 0.3 mol/L was higher than that of all the other groups,especially higher than that of the 80% ethanol pretreatment group despite that there was no significant difference among all the groups.And the test of failure mode indicated that the cohesive failure was more common,while the frequency of adhesive failure was rare in each experimental group.Conclusion:EDC pretreatment had no adverse effect on the immediate micro-shear bond strengths of Clearfil SE Bond.Meanwhile,EDC treatment did not cause more adhesive failure in immediate micro-shear test,which had further denoted what was said above.However,it needs more research to illustrate the antienzymatic role of EDC in dentin bonding.

Methods By using Microsoft Office Excel 2013 software, use frequencies, classifications of pharmacological actions and doses of TCM decoction pieces for heat clearing (DHC) from 2013 to 2015 of the First Affiliated Hospital of Xi'an Jiaotong University were analyzed. Results In our hospital, from 2013 to 2015, in top 15 DHC, the amounts of clearing away heat and toxin were more than 40%, and which of the average doses exceeded the upper limit of pharmacopoeia dosage, were more than 30%. The part that overdoes frequencies greater than 30% were 11.0% of the total, nevertheless, the part that not exceeded was 39.0% of the total. Conclusion The use of medicine for heat clearing should be carefully dialectical, paying attention to the rationality of medication. The dose should be careful and effectively regulated.

OBJECTIVE:To provide reference for further promoting the implementation ofseparation of prescribing and dis-pensingpolicy which fit China's national conditions. METHODS:Based on literature review,informant interview,and filed in-vestigation the development ofseparation of prescribing and dispensingwere compared between typical eastern and western coun-tries. Constraints ofseparation of prescribing and dispensingin China were explored,international successful experiences were summarized to inspire appropriate implementation of this policy in China. RESULTS & CONCLUSIONS:There are four con-straints in the implementation ofseparation of prescribing and dispensingin China. Firstly,the legal foundation is yet established for the professional development of pharmacists. Secondly,the medicine pricing and hospital financing systems are unreasonable. Thirdly,patients'ownership of prescriptions and right of dispensing options are monopolized by hospitals. Fourthly,safety of medi-cines use is yet ensured.Separation of prescribing and dispensingshould follow the rules of social and economic development and should be adjusted to adapt local conditions.Separation of prescribing and dispensingpolicy itself may not help to achieve the objectives of cost containment and rational use of medicines. To achieve the intended objectives,legislation of Pharmacist Law should also be accelerated to clearly define the role of pharmacist and guide the behavior of prescribers with economic levers.

ABSTRACT

Objective To investigate the preventive and therapeutic effect and mechanism of simvastatin on secondary inflammatory damage of rats with cerebral hemorrhage. Methods Sixty SD rat aged 9–12 weeks were chosen and divided into the control group, model group and simvastatin-treated group randomly with 20 rats in each group. Rats in the model group and simvastatin-treated group were infused with autologous fresh uncoagulated blood to the right brain tissue of the basal ganglia to build the cerebral hemorrhage model, while rats in the control group were treated with the same amount of normal saline. Then, rats in the simvastatin-treated group were given a gavage of 3 mg/kg of simvastatin once a day after modeling. Rats in the three groups were given nerve dysfunction score (NDS) and wet-dry weighting method was used to detect the brain water content (BWC) of brain tissues around the lesion of the rats. Then Nissl staining was conducted and the undamaged neurons were counted. Immunohistochemical SP method was applied to count the number of NF-κB, TLR4 and IL-1β positive cells in brain tissues around the lesions, and the immuno fluorescence method was employed to determine the expression levels of NF-κB, TLR4 and IL-1β proteins. Results The NDS results of the simvastatin-treated group at all time points were all significantly higher than those of the model group (P < 0.05); the BWC values of the simvastatin-treated group at all time points were all significantly lower than those of the model group at the same periods (P < 0.05); the number of the undamaged neurons around the lesions of the simvastatin-treated group at all time points were all significantly higher than those of the model group (P < 0.05); seven days after treatment, the number of the NF-κB, TLR4 and IL-1β positive cells in brain tissues around the lesions of the simvastatin-treated group were all significantly lower than those of the model group (P < 0.05), and its expression levels of NF-κB, TLR4 and IL-1β protein were also significantly lower than those of the model group (P < 0.05). Conclusions Simvastatin can inhibit the expressions of NF-κB, TLR4 and IL-1β proteins in rats with cerebral hemorrhage, and protect neurons and reduce secondary inflammatory damages by down-regulating the above protein-mediated inflammatory responses.

ABSTRACT

Objective:To investigate the clinical significance of the changes of T lymphocyte subsets in the peripheral blood of patients with gastric cancer. Methods:The lymphocyte subsets in 67 patients with gastric cancer were analyzed with flow cytometry. CEA and CA199 from all patients were also detected. Results were statistically analyzed, and the patients were followed up. Results:The aver-age values of Tc (CD8+CD28+) in patients with stagesⅢandⅣgastric cancer were 12.3±5.1 and 9.9±4.6, respectively, with significant difference (P=0.043). The average values of Tc (CD8+CD28+) in patients with elevated CEA and with normal CEA were 9.3±3.6 and 12.1± 5.6, respectively (P=0.025). Similarly, the average values of Tc (CD8+CD28+) in patients with elevated CA199 and with normal CEA were 9.3±4.2 and 12.1±5.3, respectively (P=0.029). The overall survival of patients with decreased Tc (CD8+CD28+) was lower than that of patients with normal Tc (CD8+CD28+) (8.1 vs. 15.1 months, P=0.008). Conclusion:The cellular immune function of patients with gastric cancer gradually decreased with the progression of TNM. Higher CA199 and CEA values indicated lower average values of Tc (CD8+CD28+). The overall survival of patients with decreased Tc (CD8+CD28+) was lower.

ABSTRACT

<p><b>OBJECTIVE</b>Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) resistance greatly limits the clinical therapeutic efficacy of TRAIL. Elucidating the molecular mechanism underlying TRAIL resistance will be fundamental to resolving this problem.</p><p><b>METHODS</b>Nuclear and cytoplasmic protein extraction and immuno?uorescence (IF) assay were used to detect changes in heterogeneous nuclear ribonucleoprotein K (hnRNPK) localization in H1299 cells. The evaluation of cell apoptosis in cells transfected with GFP-hnRNPK, GFP-hnRNPK S284/353A, or GFP-hnRNPK S284/353D mutant was performed using cleaved caspase-3 antibody. The gene expression of XIAP was tested by quantitative RT-PCR.</p><p><b>RESULTS</b>Previously, we reported that hnRNPK antagonized TRAIL-induced apoptosis through inhibition of PKC-mediated GSK3β phosphorylation. In this study, we further demonstrate that TRAIL treatment induces cytoplasmic accumulation of hnRNPK in H1299 cells. The hnRNPK localized in the cytoplasm has a higher capacity to antagonize TRAIL-induced apoptosis. Both ERK1/2 signaling inhibitor U0126 and ERK-phosphoacceptor-site mutant (GFP-hnRNPK S284/353A) diminish cytoplasmic accumulation of hnRNPK induced by TRAIL. Moreover, we show that XIAP is involved in hnRNPK-mediated TRAIL resistance in H1299 cells.</p><p><b>CONCLUSION</b>Taken together, these results give new insights into the understanding of the molecular mechanism associated with TRAIL resistance in lung adenocarcinoma.</p>
